Accuracy of quantitative polymerase chain reaction in samples of frozen and paraffin-embedded healthy skin for the diagnosis of canine visceral leishmaniasis Arq. Bras. Med. Vet. Zootec.
Campos,M.P.; Madeira,M.F.; Silva,D.A.; Solcà,M.S.; Espíndola,O.M.; Mendes Júnior,A.A.V.; Figueiredo,F.B..
ABSTRACT The purpose of the present work was to evaluate the accuracy of quantitative polymerase chain reaction (qPCR) performed on samples of fresh frozen tissue (FT) and formalin-fixed, paraffin-embedded (FFPE) healthy skin. This is a validation study conducted with samples from 46 dogs from an endemic area in Brazil. After sample collection, DNA extractions were conducted using commercial kits and qPCR was oriented to kinetoplast DNA (kDNA) targets of the Leishmania infantum species. The results obtained for the FFPE samples showed 63.6% sensitivity and 77.1% specificity, whereas those obtained for the FT samples showed 100% and 48.6%, respectively. Poor agreement was observed for the results of the qPCR technique with FT and FFPE samples. Our results...

Can vaccines against canine visceral leishmaniasis interfere with the serological diagnostics recommended by the Brazilian Ministry of Health? Ciência Rural
Campos,Monique Paiva de; Luca,Paula Mello De; Renzetti,Alinne Rangel dos Santos; Souza,Sara Maria Marques de; Mendes Júnior,Artur Augusto Velho; Barros,Renata Simões; Figueiredo,Fabiano Borges.
ABSTRACT: The objective of the current research was to assess seroconversion in dogs immunized with Leishmune® and Leish Tec® vaccines using rapid chromatographic immunoassay DPP® (Dual Path Platform) (DPP CVL) and enzyme immunoassay (EIE) up to one year after the vaccination protocol. The study sample comprised 28 dogs divided into two groups, each group immunized with an anti-CVL vaccine and clinically monitored for one year through clinical evaluation and laboratory tests. 22 (78.5%) dog were monitored. During the evaluation time (T1-30 days, T2-6 months, and T3-1 year after vaccination) the results for all dogs were negative for CVL, except for one animal vaccinated with Leish tec® that seroconverted in the DPP CVL test at T2. Subsequent examinations...

Correlation between chronic inflammation, immunostaining and parasite load in the genital system of female dogs naturally infected with Leishmania infantum Ciência Rural
Oliveira,Vinícius Vasconcelos Gomes de; Macedo,Simone Regina Barros de; Araújo,Flábio Ribeiro de; Ramos,Carlos Alberto do Nascimento; Ramos,Rafael Antonio Nascimento; Maia,Frederico Celso Lyra; Alves,Leucio Câmara; Silva Junior,Valdemiro Amaro da.
ABSTRACT: The aim of the present study was to investigate the correlations among chronic inflammatory reaction, immunostaining and parasite load in the genital system of female dogs naturally infected with Leishmania infantum . Animals (n = 10) used in this study were from the Department of Vector Control and Animal Surveillance of the municipality of Caruaru, state of Pernambuco, Brazil. Fragments of the vulva, vagina, cervix, uterine body, uterine horns and ovaries were submitted to histopathological analysis, immunohistochemistry (IHC) and DNA detection of amastigotes by qPCR. Correlations were found between the IHC findings and chronic inflammatory infiltrate related to L. Infantum only in the vulva and vagina; whereas, the same inflammatory reactions...

Evaluation of the presence of Leishmania infantum in bone stored in glycerol or sterilized by autoclave for cortical bone grafting Arq. Bras. Med. Vet. Zootec.
Lopes,M.A.M.; Silva,S.O.; Amorim,I.F.G.; Tafuri,W.L.; Melo,M.N.; Rezende,C.M.F..
ABSTRACT This study was performed to evaluate the effects of autoclaving and storage in 85% glycerol on cortical bone from dogs infected with Leishmania sp. We used 42 cadavers with leishmaniasis. The dogs were evaluated for the presence of Leishmania sp. in culture of bone marrow and by culturing cortical bone. From the infected animals, we harvested 42 diaphysis of the right femur for culture of cortical bone before and after autoclaving or storage in glycerol. There was no significant difference in growth of the parasite in culture of bone marrow or cortical bone. There was no growth of Leishmania sp. in culture of samples after autoclaving or storage in glycerol. Both treatments were effective in preventing the growth of the parasite in vitro, so it...

Histopathological evaluation and parasite quantification (qPCR) in the male dog's genital system after natural infection with Leishmania infantum Ciência Rural
Oliveira,Vinícius Vasconcelos Gomes de; Macedo,Simone Regina Barros de; Araújo,Flábio Ribeiro de; Ramos,Carlos Alberto do Nascimento; Ramos,Rafael Antonio Nascimento; Maia,Frederico Celso Lyra; Alves,Leucio Câmara; Silva Junior,Valdemiro Amaro da.
ABSTRACT: Canine visceral leishmaniasisis an important disease caused by the protozoon Leishmania infantum which affects several organs and systems, including the genital tract. The L. infantum tropism to the male genital system and correlation among parasite load, immunohistochemistry detection and structural changes in these organs is controversy. Therefore, the aim of this study was to evaluate this correlation in the genital organs of the male dogs naturally infected with L. infantum. Samples from testicles, epididymis, prostate, glans penis, prepuce and scrotum were collected from 19 positive adult dogs. Structural changes were observed in the testicles (5.2%), epididymis (2.6%), prepuce (5.2%) and scrotum (5.2%) of the samples positive at...

Improving methods for epidemiological control of canine visceral leishmaniasis based on a mathematical model. Impact on the incidence of the canine and human disease Anais da ABC (AABC)
Palatnik-de-Sousa,Clarisa B.; Batista-de-Melo,Leopoldina M.; Borja-Cabrera,Gulnara P.; Palatnik,Marcos; Lavor,Carlile C..
The mathematical model described by Dye (1996) condemned the epidemiological canine visceral leishma-niasis control campaign, considering it non-efficient. Using this model, we mathematically demonstrate that the control is not efficient, only at low values (rate at which latent and infectious dogs are lost by the destruction program) which match the canine seropositivity observed in the field by the immunofluorescency (IF) blood eluates analysis. With higher k values, corresponding to IF (k = 0.07) or ELISA ( k = 0.25) results in sera samples, the number of infectious dogs declines to a Ro =1 or Ro =0, respectively, interrupting the transmission and the advancement of epidemics. We also experimentally demonstrate that the dog removal, following the...
